The QKGEN® double-stranded DNA fragmentation kit uses time-dependent fragmentation enzymes that can cut double-stranded DNA into 50-1,000 bp fragments depending on the time of action. The enzyme system of this kit is composed of two enzymes, one enzyme randomly creates a cut on double-stranded DNA, and the other enzyme can recognize the cut and cut on the opposite strand to generate double-stranded DNA fragments. The resulting DNA fragment contains extremely short protruding ends, with 5´ ends containing phosphate groups and 3´ ends containing hydroxyl groups. After library preparation and sequencing, the YLGENE™ double-stranded DNA fragmentation reagent has random cutting properties. Comparing genomic DNA libraries prepared by this enzyme with those prepared by mechanical method, there was no obvious bias and no difference in sequencing coverage.
